地不容毛状根的培养

摘要: 以R10OO,15834,1855,8196等4种发根农杆菌菌株,在不同条件下,感染地不容(Stephania delavayi Diels),结果在光照条件下,R1000 诱导植物幼嫩组织产生毛状根。毛状根在MS液体培养基中生长 42 d后,趋于稳定;异喹啉生物碱含量在第 28 d时达到峰值 1104ug/gFW。6种培养基中,毛状根生长及异喹啉生物碱含量的差异很大,在 B5培养基中毛状根生长最快,Nitsch培养基中毛状根的异喹啉生物碱含量最高。B5培养基中,苯丙氨酸抑制毛状根的生长及异喹琳生物碱合成。

Abstract: On different conditions, Stephania delavayi was infected by four kinds of Agrobacterium rhizogenes strains R1000, 15834, 1855, 8196, and hairy roots were induced by the strain R1000 in light from young plant tissues. In MS liquid medium, the growth of hairy roots tended to be stable after they had been cultured for 42 days. The content of isoquinoline alkaloid came up to the summit on the 28th day, which was 1104pg / gFW. Both growth and isoquinoline alkaloid content of hairy roots were very different in six kinds of media. Among different media, hairy roots grew fastest in Bs medium, and the content of isoquinoline alkaloids was the highest in Nitsch medium.Phenylalanine inhibited both biosynthesis of isoquinoline alkaloids and growth of the hairy roots in B5 medium.
